Above the bar — 10/11 HBeAg-negative patients with pgRNA loss; first HBeAg-positive patient became undetectable
Precision BioSciences’ clinical data strengthen the PBGENE-HBV cure story with durable pgRNA loss, including its first HBeAg-positive patient.
Precision is a clinical-stage gene-editing company advancing two lead programs: PBGENE-HBV for chronic hepatitis B and PBGENE-DMD for Duchenne muscular dystrophy. PBGENE-HBV was already the company’s main clinical proof point after May 2026 data showed biopsy evidence of cccDNA reduction and durable pgRNA loss, so this filing is an expansion of an existing story rather than a surprise from nowhere.
The HBV efficacy signal is becoming broader and more durable. Of 11 HBeAg-negative patients with detectable pgRNA at baseline, 10 now have undetectable pgRNA, with average durability of five months; the first six patients with pgRNA loss have passed six months. The filing states that “10 currently have undetectable pgRNA after PBGENE-HBV treatment.” 〔0〕
| Measure (filing section) | Current | Comparison |
|---|---|---|
| HBeAg-negative patients with baseline detectable pgRNA (Results) | 10/11 | 5-month average durability |
| Patients with pgRNA loss past milestone (Results) | 6 | >6 months |
| First HBeAg-positive patient pgRNA (Results) | undetectable | 5,750 copies/mL baseline |
| cccDNA-derived transcripts (Results) | 1-log reduction | 2 administrations at 0.4 mg/kg |
The most important incremental news is the HBeAg-positive result. This population carries higher cccDNA and pgRNA burdens, so undetectable pgRNA after treatment suggests the mechanism may not be confined to patients with relatively lower viral reservoirs. The filing says, “In the first HBeAg-positive patient, treated at 0.4 mg/kg, pgRNA decreased from 5,750 copies/mL at baseline to undetectable.”
This is ahead of the early clinical bar, but not yet a clinical cure demonstration. The data reinforce the biological mechanism—cccDNA-derived transcripts fell 10-fold in one patient and pgRNA remained undetectable in most evaluable patients—but the sample remains small, follow-up is incomplete, and the filing does not provide a detailed dose-limiting-toxicity or full safety breakdown. The prior direction was already known from the May EASL update, which reported pgRNA loss in all six evaluable patients, making today’s release more about breadth and durability than a wholly new thesis.
Bottom line: PBGENE-HBV is strengthening its proof-of-mechanism story and has now reached a more demanding HBV subgroup. That is a modest step forward for the program, but the evidence is still early and too small to rewrite the overall company story.
