Fragile X IND cleared — FDA clearance; planned Phase 2a in approximately 60 adult males
Anavex’s FDA clearance opens its Fragile X clinical program, moving blarcamesine from preclinical support into a planned Phase 2a study.
Anavex is a clinical-stage CNS biotech concentrating its lead drug, blarcamesine, across early Alzheimer’s disease, Rett syndrome, and Fragile X syndrome; before this filing, it had said an FXS IND submission was expected in September 2026. This advances the Fragile X program from preparation into an FDA-authorized clinical pathway. The FDA notified Anavex that it could proceed, and the company said, “The IND is now open, enabling clinical investigation of blarcamesine in FXS in the United States.” 〔0〕 That is a real regulatory milestone, but the direction was partly anticipated because the company had already announced its intention to submit the IND in September.
The next test is clinical, not regulatory. Anavex now describes a planned randomized, placebo-controlled Phase 2a study of oral blarcamesine in approximately 60 adult males, with safety, tolerability, pharmacokinetics, behavioral, and cognitive measures. The supporting evidence remains preclinical: the release cites dose-related EEG improvements after 14 days in a mouse model, while explicitly stating that blarcamesine has not been approved and that its safety and efficacy in FXS are unestablished. 〔1〕 〔2〕
Bottom line: This is a modestly positive execution milestone that opens a new clinical program, but it does not yet validate efficacy or materially reduce development risk. The business story has moved from planned submission to permitted testing; enrollment and human data are the meaningful next proof points.
