Eton ET-700 study cuts side effects but trails GALZIN efficacy

Below the bar — ET-700 reduced hepatic copper uptake 12% vs GALZIN 46%; more study arms planned

Eton’s ET-700 study showed a major tolerability advantage over GALZIN, but weaker copper absorption blocking pushes the program toward reformulation and more testing.

Eton is building a rare-disease platform around a growing commercial portfolio, with GALZIN already marketed for Wilson disease and ET-700 intended to improve zinc therapy’s gastrointestinal tolerability while preserving copper control. Earlier company plans called for this pilot PET study to validate ET-700 and potentially support a pivotal study in early 2027.

The safety result is genuinely encouraging, but it is not the main development hurdle. Only 1 of 12 ET-700 participants reported a treatment-related adverse event, versus 8 of 12 on GALZIN, with no serious adverse events reported. 〔0〕 That supports ET-700’s core differentiation: a more tolerable regimen that could potentially improve adherence.

Hepatic copper uptake, primary analysis (Study results)-12%46%-11%
Hepatic copper uptake, intention-to-treat analysis (Study results)-18%58%-11%

The efficacy trade-off is the problem. ET-700 reduced hepatic copper uptake by only 12% in the prespecified analysis, compared with 46% for GALZIN; the supportive analysis showed the same pattern, with GALZIN’s advantage statistically significant at p=0.023. 〔1〕 For a program previously positioned as a potential improvement on existing zinc therapy, this is below the bar needed to move directly into the next clinical stage.

The result changes the program from validation to optimization. Rather than advancing the tested formulation toward a pivotal study, Eton is now pursuing faster-release versions intended to improve zinc exposure where copper absorption occurs. 〔2〕 That keeps the opportunity alive, but adds development work and leaves the central efficacy question unresolved.

Bottom line: ET-700 found the tolerability advantage Eton wanted, but not enough copper-blocking activity to clear the study’s practical development bar. The Wilson disease program advances only conditionally, through reformulation and additional testing rather than a clean pivot toward pivotal development.
