OnKure Therapeutics, Inc. (OKUR) · Nov 4, 2026 · Earnings
In line — EPS $(0.36) vs published consensus around $(0.36)-$(0.42)
OnKure’s third-quarter earnings keep both 2027 IND submissions on schedule and introduce PANORAMA, adding disease data infrastructure without clinical proof yet.
OnKure is an early-stage biotech rebuilding around two next-generation PI3Kα programs—OKI-355 for vascular anomalies and OKI-345 for breast cancer—with both IND submissions already targeted for the first half of 2027. That roadmap was established in the March financing and reiterated in the second-quarter update, so the main question here is execution, not a newly discovered milestone.
The development plan remains on schedule, but is not materially advanced. OnKure again says both lead programs are progressing through IND-enabling work and remain targeted for 1H 2027 submissions. 〔0〕 That is reassuring execution, but largely confirmation of the prior quarter’s message rather than a new de-risking event.
PANORAMA broadens the vascular-anomalies strategy, though it is still an evidence-building project. The new registry will collect longitudinal information on disease course, treatment patterns, genetic variants and clinical characteristics, with genotyping offered to participants. 〔1〕 This could improve patient identification and future trial design, but it does not yet provide efficacy, safety or regulatory validation for OKI-355.
The quarter’s financial result is essentially in line, with spending rising as the pipeline moves toward the clinic. Reported EPS of $0.36 per share lost was around the published consensus range of roughly $0.36-$0.42, while the company gave no product-revenue update. R&D increased to $15.1 million from $11.9 million as outsourced preclinical work, manufacturing and consulting costs rose.
| Q3 2026 | Q3 2025 | Consensus | |
|---|---|---|---|
| Net loss (Financial Results) | $17.8M | $14.7M | |
| Loss per share (Financial Results) | $(0.36) | $(1.09) | $(0.36)-$(0.42) |
| R&D expense (Statements of Operations) | $15.1M | $11.9M | |
| Cash, equivalents and securities (Financial Results) | $163.3M |
Cash still supports the current plan, but the cushion is shrinking. Cash, cash equivalents and marketable securities fell from $176.4 million at June 30 to $163.3 million at September 30, while management continues to expect runway into 2029. The balance sheet therefore remains adequate for the stated near-term milestones, without eliminating the longer-term financing risk typical of a clinical-stage company.
Bottom line: This is a steady execution quarter with one useful strategic addition, not a major change in OnKure’s investment story. The company is still moving toward the same 2027 IND milestones, with PANORAMA adding infrastructure but no clinical proof yet.
