Clinical milestones advanced — Phase 2b and Phase 3 programs fully enrolled; cash runway into mid-2028
Kailera’s third-quarter earnings show multiple obesity trials advancing toward 2027 data, supported by cash into mid-2028 despite sharply higher R&D spending.
Kailera is a newly public, clinical-stage obesity biotech building a four-drug GLP-1 pipeline around injectable and oral ribupatide, with no commercial revenue and its value concentrated in upcoming clinical readouts. The prior quarter had already established the broad path: ribupatide injection’s Phase 2b trial was fully enrolled, the oral program was headed toward Phase 3 in the first half of 2027, and management expected cash to last into mid-2028.
The operating story moved forward, but mostly by confirming execution already expected. Kailera now says the ribupatide injection Phase 2b trial and the roughly 5,000-patient KaiNETIC Phase 3 program are fully enrolled, safiglipron’s Phase 2 trial is also fully enrolled, the FDA has completed its End-of-Phase 2 meeting for oral ribupatide, and KAI-4729’s IND is active. The oral ribupatide Phase 3 start remains targeted for the first half of 2027. 〔0〕
The important change is a denser, more visible 2027 catalyst calendar—not a new clinical proof point. Mid-2027 now carries expected data from the high-dose ribupatide injection Phase 2b trial and safiglipron Phase 2, while KAI-4729 is targeted to enter Phase 1 by year-end 2026; KaiNETIC Phase 3 data remain a mid-2028 event. The milestones strengthen the execution narrative, but they are timelines and enrollment updates, not efficacy results. 〔1〕
The balance sheet still supports the plan, although the burn rate is rising sharply. Cash, cash equivalents and marketable securities totaled $1.04 billion and management still expects funding into mid-2028. But quarterly R&D expense rose to $84.7 million from $27.0 million, while net loss widened to $95.7 million from $39.6 million as the portfolio moved deeper into clinical development.
| Q3 2026 | Q3 2025 | Dec. 31, 2025 | |
|---|---|---|---|
| R&D expense (Statements of Operations) | $84.7M (+213.6%) | $27.0M | |
| G&A expense (Statements of Operations) | $21.7M (+65.2%) | $13.1M | |
| Net loss (Statements of Operations) | $95.7M (+141.7%) | $39.6M | |
| Cash, equivalents & marketable securities (Financial Results) | $1,037.6M | $652.7M |
Bottom line: Kailera is executing against the obesity development plan and has enough cash to reach the next major milestones, but this quarter adds no clinical efficacy evidence and shows the cost of advancing several programs at once. The update is modestly constructive rather than story-changing.
