Spyre earnings miss as IBD advances, rheumatic bets retreat

Miss — Common EPS $(0.94) vs published consensus around $(0.78)

Spyre’s Q3 earnings missed consensus as IBD programs advanced, while failed rheumatic endpoints narrowed SPY072’s path and funding shifted toward HS.

Spyre is a clinical-stage immunology biotech building long-acting antibody combinations in IBD while expanding its TL1A platform into dermatology, especially hidradenitis suppurativa; the prior setup already included positive monotherapy data, the October equity raise, and a scheduled 2026 rheumatology update.

The quarter missed the published EPS bar. Common-stock loss was $0.94 per share versus a published consensus around $0.78, while there is no product revenue to offset the clinical spending. R&D expense nearly doubled year over year as manufacturing and trial costs ramped, so the miss reflects a more expensive development engine rather than a revenue shortfall.

The core IBD thesis advanced, but mostly along an already-known path. Spyre completed proof-of-concept for all three SKYLINE monotherapies, and faster-than-expected Part B enrollment allows a larger combination study with topline results still targeted for the third quarter of 2027. 〔0〕 That strengthens the company’s main combination strategy, but the positive SPY003 result was announced in September and the broader SKYLINE plan was already public, making this more confirmation and execution detail than a new thesis reset.

SPY072’s rheumatology expansion was cut back sharply. RA showed clinical activity but fell below Spyre’s internal bar for advancement, while PsA and axSpA failed their primary endpoints; the company is abandoning rheumatic-disease development for SPY072. 〔1〕 This narrows the asset’s opportunity, though it also concentrates resources on the more important IBD and HS programs rather than funding a weak indication.

The new HS push is financed, not yet validated. Spyre has enough cash and securities to support operations into the second half of 2029 on a pro forma basis, including October financing proceeds, and plans to take SPY072 into pivotal HS development. The funding removes a near-term financing constraint, but the HS program remains an execution and clinical-risk story; its positive rationale comes from external proof-of-concept rather than a Spyre pivotal readout.

Bottom line: This is a mixed quarter that leans negative: the main IBD platform is progressing, but the EPS miss and failed rheumatology expansion show a costlier, narrower development story. The added HS funding preserves momentum, but does not yet replace the lost rheumatology upside with clinical proof.
